潜在性抗肿瘤和抗炎症靶点PARP14的研究进展

【摘要】 PARP14是聚ADP-核糖聚合酶的超家族成员中一种单ADP-核糖转移酶。PARP14通过对靶蛋白的ADP-核糖基化修饰[poly(ADP-ribosylation),PARs],参与着广泛的体内生理过程,在细胞的转录调控、应激反应、DNA修复、RNA剪切、线粒体功能、分裂和核小体的形成过程中都发挥着重要作用。PARP14与癌症(如肝细胞癌、多发性骨髓瘤和胰腺癌)、代谢性疾病以及炎症性疾病的发生、发展密切相关,使其成为极具潜力的药物研发靶点。该文综述了PARP14的结构和生物学功能,并总结了PARP14在疾病中的作用以及现有的PARP14小分子抑制剂和降解剂,对PARP14抑制剂和降解剂的研发提供一个简明的更新,协助选择性PARP14抑制剂和降解剂的发展,为靶向PARP14的药物或化学增敏剂的研发提供参考。

【Abstract】 PARP14 is an intracellular single ADP-ribose transferase member of the superfamily of polyADP-ribose polymerases. PARP14 is associated with a wide range of biochemical transformations in vivo through poly(ADP-ribosylation, PARs) modification of target proteins. For example, it plays an important part in cellular transcriptional regulation, stress response, DNA repair, RNA splicing, mitochondrial function, division, and nucleosome formation. PARP14 is closely related to the development and progression of cancers(such as hepatocellular carcinoma, multiple myeloma and pancreatic cancer), metabolic diseases, and inflammatory diseases, making it a potential drug discovery target. This research reviews the structure and biological functions of PARP14, and summarizes the role of PARP14 in disease, as well as the existing PARP14 small molecule inhibitors and decompressors. It provides a brief update to the research and development of PARP14 inhibitors and decompressors to assist in the development of selective PARP14 inhibitors and decompressors. It provides reference for the research and development of drugs or chemical sensitizers targeting PARP14.
