普瑞巴林胃漂浮缓释片的制备及体外释放研究

【摘要】 目的制备普瑞巴林胃漂浮缓释片,并通过紫外分光光度法考察其体外释放影响因素。方法以普瑞巴林为模型药物,分别以聚氧乙烯303、聚氧乙烯N-60K、羟丙基甲基纤维素K15M和海藻酸钠为基质制备胃漂浮缓释片,采用紫外可见分光光度法,在波长365 nm处测定吸光度,计算释放度。结果聚氧乙烯303每片用量为250 mg、聚氧乙烯N-60K每片用量为250 mg和海藻酸钠每片用量为150 mg时漂浮性能和释放度最佳。结论制备的普瑞巴林胃漂浮缓释片漂浮性良好,且能够达到缓释的目的。

【Abstract】 Objective To prepare the pregabalin gastric floating sustained-release tablets and investigate the drug release by UV. Methods Pregabalin was used as a model drug with Polyox WSR 303, Polyox WSR N-60 K, HPMC K15 M and sodium alginate as matrix materials. The content of pregabalin was determined by ultraviolet spectrophotometry at 365 nm and accumulative release amount was calculated. Results The tablets showed a good sustained-release and continuous floating property with Polyox WSR 303 250 mg/tablet, Polyox WSR N-60 K 250 mg/tablet, sodium alginate150 mg/tablet. Conclusions Floating state of pregabalin gastric floating sustained-release tablets was maintained during the whole release process, and sustained-release properties was achieved.
